Keeping it together: How single-molecule imaging can reveal the mechanisms of DNA double strand break repair

DNA double strand breaks (DSBs) are an extremely toxic form of DNA damage and nature has developed elaborate mechanisms to repair them. In humans, the majority of DSBs are repaired by the non-homologous end joining (NHEJ) pathway. During NHEJ, a multi-protein complex is assembled on DNA, which pairs the two DNA ends and ultimately rejoins them. In this talk, I will describe how my laboratory is using novel single-molecule imaging approaches to probe the mechanism of NHEJ with increasing molecular resolution.
